    Application of extreme environmental conditions to resuscitation of viable but non culturable E. coli DH5α

    The resuscitation of Viable But Non-Culturable (VBNC) state in Escherichia coli DH5α as one of the most usable expression host was investigated. The VBNC state in bacteria is defined as while the cells are alive but unable to grow visibly on nonselective growth medium. After collecting several samples, Non-culturable E. coli DH5α (that has undertaken on various recombinant manipulations) were divided into different groups in order to carry-out different experiments. They were treated to heat shock at 42ºC in different periods of time, different concentration of Bile-salts and NaCl and combinational of these methods. The results showed that the almost of resuscitation treatment had positive effect on reactivation of VBNC E. coli DH5α. The combination of these parameters (various NaCl and bile salts concentration and heat shock method at 42 °C in different time) in a binary manner, also inferred to suitable results. Furthermore, by applying the three stresses simultaneously we achieved optical density up to 0.58 and 9×108 CFU/ml which had presented the best results. The results show that by applying some alterations in the condition of such recombinant E. coli DH5α, the growth path of these bacteria which remain to a VBNC phase can be changed to the normal status

    Alleviation of experimental allergic encephalomyelitis in C57BL/6 mice by Soy Daidzein

    Experimental allergic encephalomyelitis (EAE) is considered as the murine model of multiple sclerosis. Daidzein a phytostrogenic compound of soy is known to impose immunomodulatory and antioxidative effects. We conducted this study to assess the potential protective and therapeutic effects of daidzein on allergic encephalomyelitis. C57BL/6 mice were induced with allergic encephalomyelitis using myelin oligodendrocyte glycoprotein (35-55) and received daidzein or dimethyl sulfoxide as the vehicle control. To assess the protective effect of daidzein, the mice were administered with 20 mg/kg of daidzein from 21 days prior to 21 days post EAE induction on a daily basis. To evaluate the therapeutic effect of daidzein, mice were fed with 300 mg/kg daidzein after the appearance of the first clinical signs for 10 days. One day after the last gavage, the mice were sacrificed. Spleen and brain were removed for further histological and immunological analysis. Feeding mice with low dose of daidzein prior to disease induction did not affect disease severity. However, treating with high dose of daidzein after the onset of the disease reduced interferon-3 and interleukin-12 secretion, enhanced interleukin-10 production, suppressed lymphocyte proliferation, and decreased cytotoxicity as judged by lactate dehydrogenase release. In conclusion, daidzein reduced the extent of demyelination and disease severity. Chronic oral therapy with low dose of daidzein did not prevent experimental autoimmune encephalomyelitis.     Effects of intermittent fasting on experimental autoimune encephalomyelitis in C57BL/6 mice

    Several religions recommend periods of fasting. One of the most frequently asked questions of MS patients before the holy month of Ramadan is weather fasting might have an unfavorable effect on their disease course. This debate became more challenging after the publication of experimental studies suggesting that calorie restriction prior to disease induction attenuates disease severity. We conducted this study to assess early and late effects of fasting on the animal model of MS, known as autoimmune encephalomyelitis. EAE was induced in the C57BL/6 mice, using Myelin Oligodendrocyte Glycopeptide (MOG) 35-55 and they fasted every other day either after the appearance of the first clinical sign or 30 days after disease induction for ten days. Thereafter, the mice were sacrificed for further histological and immunological evaluations. Intermittent fasting after the establishment of EAE did not have any unfavorable effect on the course of disease. Moreover, fasting at the early phase of disease alleviated EAE severity by ameliorating spinal cord demyelination. Fasting suppressed the secretion of IFN-γ, TNF-α and raised IL-10 production in splenocytes. Fasting was also associated with a lower percent of cytotoxicity. Intermittent fasting not only had no unfavorable effect on EAE but also reduced EAE severity if started at early phase of disease. © Summer 2016, Iran J Allergy Asthma Immunol. All rights reserved

    Identification and molecular analysis of mercury resistant bacteria in Kor River, Iran

    Mercury (Hg) is one of the most important toxic pollutants widespread in the environment. It is being extensively used in industrial applications (chlor-alkali electrolysis, fungicides, disinfectants, dental products, etc), resulting in local hot spots of pollution and serious effects on biota and humans. The aim of this study was to identify mercury resistant bacteria and extract their plasmids and DNA. In this study, mercury-resistant bacteria were isolated and characterized from mercury-polluted sediments in Kor River in Iran. The samples were cultured in different media cultures, identified using biochemical tests, and due to the relationship between antibiotic and mercury resistance, they were isolated based on these two factors. The plasmids and DNA were extracted from the most resistant bacteria to both antibiotic and mercury and the sizes were determined using agarose gel electrophoresis. A 12.3 Kb plasmid from Serattia sp. and Escherichia coli and using Sau3A1 enzyme, some DNA fragments (4, 6, 8 and 10 Kb) from Pseudomonas sp., Serattia sp. and Escherichia coli were also extracted.Key words: Mercury, resistant, bacteria, DNA, plasmid extraction, restriction endonuclease

    Effect of oral genistein administration in early and late phases of allergic encephalomyelitis

    Objective(s): Experimental allergic encephalomyelitis (EAE) is an autoimmune disease validated as animal model of multiple sclerosis (MS). Administration of genistein, a phytoestrogenic component of soy, to mice at the onset of EAE is known to attenuate the clinical signs of the disease. The potential effects of genistein on established EAE is less studied. In the current study, we aimed to compare the effects of genistein administration on EAE severity in early and late phases of the disease. Materials and Methods: The C57BL/6 mice were induced with EAE, using MOG 35-55 and gavaged with genistein (300 mg/kg) either after the appearance of the first clinical sign or 30 days post disease induction for ten days. 24 hr after the last gavage, mice were sacrificed. Brains and spleens were removed for assessing lymphocyte proliferation, cell cytotoxicity, and cytokine profile. Spinal cords were dissected to assess the amount of demyelination using Luxol fast blue/cresyl violet staining. Results: Administering mice with genistein, after the establishment of EAE, did not reverse the clinical signs of disease. However, treating with genistein at the onset of disease alleviated the clinical signs by reducing neuronal demyelination. Genistein suppressed the production of IFN-γ and enhanced IL-10 secretion in splenocyte and brain. Genistein also reduced IL-12 and TNF-α secretion in splenocytes, suppressed the proliferation of T-cells, and reduced the cell cytotoxicity. Conclusion: Genistein oral therapy might only reduce EAE severity if started in early phases of the disease

    Using Ferula assafoetida essential oil as adult carob moth repellent in Qom pomegranate orchards (Iran)

    Carob moth, Ectomyelois ceratoniae (Lep.: Pyralidae), is the key pest of pomegranate fruits in most countries like Iran. Chemical insecticides cannot be used for the control of carob moth because they have biological and behavioral traits. Other control methods have no sufficient efficiency. So it is necessary to use new methods. Some pest control management such as application of semiochemicals like repellents are common practices. Ferula assafoetida essential oil is a botanical material and itsrepellency on some pests is proved under laboratory conditions. In this research, its repellency was studied on adult carob moth, in Qom pomegranate orchards, during a two farming seasons study (2007- 2008). Two orchards (treatment and control) were selected in about 3 km distances from each other. In treatment and control plots, F. assafoetida essential oil and ethanol (solvent) were used, respectively. During the farming season, number of rotten pomegranates of treatment and control trees were counted and removed weekly. The percentage of rotten pomegranates on trees was calculated at the end of the season. Two sample t-test showed significant differences between weekly (T = 2.22, df = 38, P = 0.032), (T = 3.11, df = 58, P = 0.003) and end of the season (T = 2.97, df = 16, P = 0.012), (T = 3.54, df = 48, P = 0.001) data of treatment and control orchards in 2007 and 2008, respectively. Number of rotten pomegranates of treatments was lower than control. Comparison of treatment and control orchards data showed damage lessening in treatment plot. Laboratory results proved repellency effect of F.assafoetida essential oil on adult carob moth in orchard conditions.Key words: Carob moth, semiochemicals, Ferula assafoetida essential oil, repellency, Iran
